What is TB-500?

TB-500 is a peptide consisting of 43 amino acids that is best known for its remarkable healing properties.

It is the synthetic version of the naturally occurring protein Thymosin Beta-4, which is encoded by the TMSB4X gene and is found in both human and animal cells.


Thymosin Beta-4 promotes a variety of essential biological functions such as [1]:

  • Lowered inflammation

  • Growth of new blood cells

  • Increased production of immune cells

  • Faster wound repair

Since Thymosin Beta-4 is naturally produced by the thymus gland in response to injury [2], people logically conclude that injecting it can accelerate the healing process.

How does TB-500 work, you may ask?

The main mechanism of action in TB-500 involves the upregulation of a cellular protein called “Actin”.

Actin plays a vital role in keeping the structural and functional integrity of cells intact, especially in in muscle cells. This means TB-500 may lead to the growth of new cells and heal tissue injuries, while improving muscle flexibility.

My “test subject” has personally noted the potential power of TB-500 in treating injuries within joints, tendons and ligaments as well.

Due to the low molecular weight of TB-500, it is extremely mobile.

This is drastically overstating the science behind how it works, but TB-500 can basically “find” the parts of your body that are injured and selectively upregulate actin at the site of injury.

No matter where it is injected, it can travel through the body without getting “blocked” or stuck anywhere.

For this reason alone, TB-500 is very popular as a doping drug in race horses [3]. And it’s so effective that active efforts are being made to completely ban its use [4]:

Australia’s strict drug-free racing policy would result in a minimum period of disqualification of two years for any person found guilty of injecting TB-500 into a racehorse.

“We have been aware of this substance for a number of years and we are confident we have the strategies and testing capabilities to control it. “This drug has no place in racing, its use is easily detected in the thoroughbred and its administration to a racehorse would be viewed very seriously.”

Naturally, athletes have taken an interest towards using TB-500 for their own athletic endeavours.

We can’t say for sure what its effect on humans will be, but sports authorities aren’t taking any chances [5]:

“Indeed, TB-500 packages have recently been confiscated by the Belgian Customs during a routine control.

The case has received worldwide attention from the media since it was connected to a professional athlete which was still employed by his team, who declared that the product was for personal use.

In another case, a former team doctor was arrested in possession of the substance.

Outside of doping, there are very few scientifically validated reports of TB-500 use in humans.

The earliest one involves a 5-year-old girl who received a TB-500 injection in 1974 because she had a dysfunctional thymus gland [6].


And the number of human clinical trials involving TB-500 can be counted with two hands.

 

So what could a TB-500 dosage potentially do for a human?

 

Before I give my best guess at the answer, I want to be very clear: TB-500 is NOT a therapeutic peptide marketed for human consumption.

 

If you try to buy it, you’ll see it’s targeted primarily towards research and animal use.

 

This is because virtually every study touting TB-500’s health benefits are done in animals and cell cultures.

 

While some biohackers and bodybuilders have experimented with the drug, their results are speculative and anecdotal at best.

TB-500

Benefits of Thymosin Beta-4

Recent research studies have shown that Thymosin Beta-4 provides numerous health benefits which go beyond tissue and wound healing.

Thymosin Beta-4 Can Repair Heart Muscles

An in vivo study involving heart cells found that Thymosin Beta-4 may be effective in healing heart injuries [7].

In particular, Thymosin Beta-4 was able to stimulate vessel growth while simultaneously preventing the death of heart cells.


One of the few clinical trials involving humans was actually able to replicate this finding in patients with myocardial infarction [8]:

“The pilot study, the first human study conducted with Tβ4 in patients following a heart attack, demonstrates the potential clinical benefits of Tβ4 in repairing and regenerating damaged tissue and improving cardiac function.

This important advance not only elaborates on data generated in previous animal studies, but also reflects the need to conduct larger clinical trials with Tβ4 in these patients”

Not only were the patients’ hearts more efficient at pumping blood, but they experienced no complications whatsoever 6 months after Thymosin Beta-4 treatment.

Thymosin Beta-4 May Accelerate Healing From Traumatic Brain Injury (TBI)

A 2012 paper reported rapid results in cognitive recovery within rats who had TBI [9].

As early as 6 hours following injury, rats treated with Thymosin Beta-4 experienced functional recovery, and the creation of new brain cells AND blood vessels took place as early as 24 hours post-injury.

Thymosin Beta-4 Could Be Used To Treat Eye Problems

As early as 2007, Thymosin Beta-4 was being examined as a potential clinical solution to repair eye damage [10].

Some of the applications speculated include:

  • Diabetic Eye Disease

  • Faster healing from cataract surgery

  • Dry eyes

  • Chemical injuries

Speaking of dry eyes…A phase 2 clinical trial in humans found Thymosin Beta-4 eye drops to be effective for treating severe dry eye [11].

9 treated patients all had significantly reduced discomfort and improved tear volume production, even 28 days following treatment!

Thymosin Beta-4 Is Useful For Treating Skin Wounds That Don’t Heal

In preclinical animal studies involving various kinds of rats and mice (steroid-injected, old, diabetic), Thymosin Beta-4 was shown to be effective in healing skin wounds [12].

But wait, there’s more!


The same review I just quoted also examined two phase 2 clinical trials involving patients with ulcers.


In the patients that healed using Thymosin Beta-4, the healing process was a MONTH faster compared to the placebo.


As a side note, the authors also noted how Thymosin Beta-4 was able to inhibit further inflammation and infection.


So as an inflammatory agent, Thymosin Beta-4 shows great promise for future studies in humans.

Thymosin Beta-4 May (Or May Not) Treat Joint Inflammation

Even in animal studies, the science on this is not clear.

According to a 2017 review of Thymosin Beta-4 and rheumatoid arthritis, there are a handful of studies touting Thymosin Beta-4 exhibiting anti-inflammatory effects [13].


However, there were a fair number of reports saying Thymosin Beta-4 increases the secretion of pro-inflammatory compounds.


And while increase Thymosin Beta-4 expression is uniformly seen in the joint fluid of rheumatoid arthritis patients, it is uncertain whether this outcome is beneficial or harmful.

Thymosin Beta-4 Could Heal Ligament Injuries

Yet another rat study involving the local administration of Thymosin Beta-4 discovered that Thymosin Beta-4 has “potential clinical use in repairing ligaments” [14].

After transecting the medial collateral ligament (MCL) in rats and treating rats with Thymosin Beta-4 a month, the treated rats had significantly better biomechanics function.


Moreover, the collagen fibers within the treated rats were uniform and evenly spaced out.


This implies that Thymosin Beta-4 heals wounds and injuries in a way where normal physical movement can be restored.


You’ll notice we didn’t say anything about muscles, flexibility, stamina or endurance so far…


…and that’s because all of those supposed benefits are experimental and self-reported.


I’ll tell you all about this in a bit, but first:


What is the BEST TB-500 dosage for someone who is going to experiment with this peptide?

When it comes to choosing the TB-500 dosage that’s right for you, there are a few things you need to know about.

First, you need a handy-dandy TB-500 dosage calculator to know EXACTLY how to prepare it [15].

As peptide users already know, TB-500 comes in the form of a lyophilized powder. Therefore, it needs to be reconstituted with bacteriostatic water before it can be injected intravenously, intramuscularly, or subcutaneously.


You MUST get the math 100% right on this, otherwise the TB-500 injection will be too small or too much.

 

Second, you need to make sure you are storing your materials correctly.

 

The TB-500 powder itself should be kept in a room that is dry, cool and pitch black.

 

Once the powder is reconstituted in the vial, the vial itself needs to be stored in a refrigerator to avoid any potential degradation.

 

Third, based on how long you intend to use TB-500 for, you will need to buy enough bottles to last you for at least 3 months.

 

This way, you can see how your TB-500 dose is affecting you and whether you need to keep on taking it.

 

Lastly, it’s important to remember that everyone will respond to a TB-500 dose differently. Some may notice the effects taking place much faster or slower compared to other people.

 

And on top of that, the dose itself CAN make a big difference in what happens.

 

So can the severity of the injury/wound being treated, and one’s intentions behind using their TB-500 dose of choice. Since you’re only using a few milligrams at a time, adding or removing 1-2 mg of TB-500 can completely change the outcome.

 

Many of the TB-500 dosage recommendations you will find are entirely dependent on first-hand observation and experience of those who have already used it on themselves (or their animals).

 

Unlike other therapeutic medications, there is no “standardized dose” for TB-500 that currently exists.

 

For example, Ben Greenfield recommends a TB-500 dose of 7.66 mg per week for a 200-pound man on the basis of what’s used for greyhound dogs [16].

 

Other people will recommend 2-4 mg of TB-500 administered twice a week, evenly spaced out — Monday and Thursday, for instance.

With that being said, what’s the best TB-500 dosing strategy for someone who’s using it for the very first time?

TB-500 For Beginners

The best way to describe how beginners should approach their TB-500 dosage of choice would be to use an analogy.

If you take creatine monohydrate, you’re probably aware of the loading protocol [17]:

“…a common approach is to take 20 grams of creatine daily for 5–7 days. This dose is typically divided into four 5-gram servings throughout the day.

Research shows that this regimen can effectively boost creatine stores by 10–40%.

After the loading phase, you can maintain your creatine stores by taking a lower dose of creatine, which ranges from 2–10 grams daily”

A similar approach is recommending for people who are new to TB-500 dosing.

Here’s a rough sketch of what a TB-500 loading cycle would look like for treating an injury in an animal:

Loading Phase

  • What To Take: 1-3mg of TB-500

  • How Often To Take: 2-3 times every 2 weeks

  • For How Long: Until injury completely heals

Maintenance Phase

  • What To Take: 2-5 mg of TB-500

  • How Often To Take: 2 times per week

  • For How Long: 4-6 weeks

Put simply, you get your system accustomed to higher Thymosin Beta-4 concentrations before you transition towards a maintenance dose.

My “test subject” told me that the maintenance dose alone is useful when you want to recover faster and lower the risk of injury while training.

He also said you can push yourself harder in competitive events and “go harder for longer”.

Of course, all of this is based on HIS own observations and yours many not necessarily be the same.

Anybody who uses TB-500 dosing for recovery purposes should be well aware of how their own body responds and adjust accordingly.

Some will respond better with a low TB-500 dose, some will do well with a higher TB-500 dose, and some people will be in the middle.

TB-500 For Healing

TB-500 has shown a ton of potential in healing injuries within experimental animal models [18].

Time and time again, rat studies keep showing that Thymosin Beta-4 will accelerate wound treatment [19].


And some animal studies even put forth the idea of Thymosin Beta-4 offering rapid pain relief [20].


If an injury isn’t healing at all, or it’s taking too long, a regimen of TB-500 dosing becomes an attractive option to bodybuilders and athletes.


Here’s what my research subject noticed when using TB-500 for healing…


If you want TB-500’s normal healing powers and you’re a complete newbie, go with the lowest end of the TB-500 dosage range I just gave.


If you’re experienced and you’re doing another round of TB-500, stay in the middle.


But for an accelerated healing effect, stick with the high end of the range.


As I said before, everyone’s circumstances are completely different.


Some of us train harder than others, are more/less injured than others, and/or are taking other medications alongside TB-500.


It’s up to you to assess the effects of your TB-500 dose and whether it’s working for you.


Sure, there are hundreds of anecdotal reports of people using TB-500 and having their own success with it.


But you are not them and they are not you. Keep that in mind!

TB-500 For Muscle Growth

One supposed effect of using the right TB-500 dose is making all kinds of lean muscle gains.

Here’s a short list of some of the effects people are claiming [21]:

  • Increased endurance

  • Increased strength

  • Increased muscle growth

  • Improved flexibility of joints

  • Improved muscle tone

Because these effects are seen when TB-500 is used on race horses, it is assumed that similar effects will be seen in humans.

A plausible explanation for this involves the up regulation of actin.

As it promotes cell proliferation and healing, which are both necessary for gaining muscle, therefore TB-500 MUST build muscle!

And this is in addition to healing damaged muscles. However, these are nothing more than mere claims.

Even my “test subject” noted that TB-500 has nothing to do with getting stronger or building more muscle.

TB-500 is first and foremost a compound designed for accelerating healing and recovery.

And even though athletes WILL get banned for using TB-500 [22], there is no evidence whatsoever in animals indicating TB-500’s ability to add skeletal muscle tissue to your body.

Long story short, TB-500 is not the hot “anabolic steroid” you may have been misled to believe it is.

TB-500 For Hair Growth

A surprising claim made by people following a TB-500 dosage protocol is that it can increase hair growth.

Not only can TB-500 encourage the growth of lost hair, but it can also darken grey hair and make your current head of hair thicker.


Some will even go as far as to claim it can combat the hair loss effects of male alopecia (hair loss due to your genetics).


But as with the claims of building muscle, this is entirely anecdotal and cannot be taken seriously. And this excludes the fact that some people swear by it while others haven’t noticed any difference.


For one, if you were to actually go with this approach, you would have to inject TB-500 into your scalp. This is dangerous to do on your own without the supervision of a medical professional.


So where did this rationale come from?


Probably from 3 studies done in rats and mice where Thymosin Beta-4 induced hair growth [23, 24, 25].

Outside of that, I would not recommend the use of TB-500 for growing more hair.

And you definitely won’t build muscle or grow a new head of hair with TB-500. But if the right TB-500 dose is found, it may be beneficial for someone who needs to treat a painful injury, recovery faster, and other varied benefits.
