What is TB-500?

TB-500 is the synthetic derivative of a peptide naturally produced by your thymus called Thymosin Beta-4.

It belongs to a group of peptides called “beta-thymosins”, which are involved in essential cellular processes such as cell death, cell migration, and the formation of new blood vessels [1].

Thymosin Beta-4 can also upregulate actin, a cellular protein that is necessary for your muscles to contract [3]. However, there lies one problem with this mechanism…


Thymosin Beta-4 is only produced in small amounts, which thereby limits the speed at which the body can recover from injuries.

Which begs the question…does Thymosin Beta-4 actually live up to its promise?

Does Thymosin Beta-4 Actually Work?

It cannot be said with medical or scientific certainty if Thymosin Beta-4 actually works in helping people recover from injuries.

With that being said, many “test subjects” have anecdotally reported their results and experience with using the peptide.

Here are just some of the claims you will find:

  • The acute or chronic pain related to the injury disappears pretty quickly

  • Healing of the wound itself is much faster compared to what the body would naturally do

  • Range of motion and flexibility are restored, and improved in rare cases

Without question, an accelerated rate of recovery is always cited amongst TB-500 “test subjects”.

Other benefits such as improved strength, lean muscle gain, and hair growth are not substantiated by the reports of research subjects.

You may find 1-2 animal studies supporting these claims at best, but in practice it would be highly ineffective to use TB-500 for these purposes.

Anecdotal claims about the benefits of TB-500 are one thing, but the science may paint a completely different picture. Let’s investigate what scientists have concluded about Thymosin Beta-4 in legitimate research studies…

Thymosin Beta-4 May Treat Brain Injuries And Disorders

A comprehensive review conducted in 2015 found that Thymosin Beta-4 showed great potential as a regenerative therapy for neurological injury and degradation.

Not only can it promote neuroplasticity in the central nervous system and peripheral nervous system, but by extension it can lead to significant improvements in behavior [7].


Another study conducted in cell cultures discovered Thymosin Beta-4’s ability to stabilize the blood brain barrier, which is another pathway through which it can be beneficial for cognitive health [8].

Thymosin Beta-4 Could Help Repair Various Organ Injuries

Thymosin Beta-4 has been identified as a novel agent for treating liver diseases such as liver fibrosis, in which involves the liver becoming dysfunctional due to excessive scarring [9].

One study conducted in 2019 involved rats who were diagnosed with acute kidney injury caused by ischemia, which can be described as an inadequate supply of blood delivered to the kidney.

Thymosin Beta-4 was able to drastically reduce the damage from the injury and lower the amount of inflammation taking place [10].

 

And if that wasn’t enough, Thymosin Beta-4’s regenerative effects were also observed in strokes and myocardial infarctions [11].

Thymosin Beta-4 Shows Usefulness For Nerve Cell Damage

This was an interesting application that usually escapes the radar of most peptide enthusiasts.

A patent was announced a few years ago to treat nerve cell damage with Thymosin Beta-4, which is present in conditions such as peripheral neuropathy.


This implies that Thymosin Beta-4 may be useful for treating muscle wasting, paralysis, severe pain, and other symptoms related to peripheral neuropathy [12].

Thymosin Beta-4 Can Lower Chronic Inflammation

A review published in 2018 examined Thymosin Beta-4’s anti-inflammatory effects, and the results were promising.

Not only can Thymosin Beta-4 be used in several diseases related to inflammation, but there are multiple mechanisms through which it exerts its benefits [13].

Thymosin Beta-4 Can Heal Ligament Injuries

In rats, Thymosin Beta-4 healed ligament injuries while also restoring biomechanical movement and integrity 4 weeks after surgery was done.

 

This was evidenced by the collagen fibers being evenly spaced when the injury was fully healed [14].


The verdict is still uncertain regarding joint and bone injuries, but many anecdotal reports from my research subjects indicate that TB-500 may be effective for those purposes.

TB-500 Side Effects

Due to the lack of human clinical trials and studies, the reported TB-500 side effects that you hear about are entirely anecdotal in nature.

This means you’ll need to go through multiple bodybuilding forums and discussion groups to get a clear picture of what you should expect.

But no reports so far have found anything which will lead to significant harm with TB-500 use.

Here are what “test subjects” have claimed to experience when using TB-500:

  • Headache

  • Nausea

  • Fatigue

  • Head rush

Generally, these side effects appear to subside after the body gets used to higher levels of TB-500.

The list of TB-500 side effects presented above are making the following four assumptions:

  1. You are using a PURE bottle of TB-500

  2. You are properly storing it in a cool, dry place devoid of light

  3. You are preparing the peptide and reconstituting it in bacteriostatic water.

  4. You are injecting TB-500 using the proper protocol for safe and sterile injections

  5. You are using a moderate TB-500 dose, or at least a dose your body can appropriately handle.

Of course, this doesn’t exclude the fact that using peptides in general can potentially lead to side effects.

This includes skin irritation, increased water retention, dry mouth, or slightly elevated blood pressure [15].

 

You may have heard rumors about the link between TB-500 and cancer, but this is based on a mis-interpretation of the scientific literature.

Yes, Thymosin-Beta 4 is usually over-expressed in in organisms with cancer [16]. However, this is because Thymosin Beta-4 levels are elevated in response to the cancer so that new white blood cells can be formed.

 

The science on this is unclear in either direction, so don’t blindly believe what you read on the Internet.

 

So what can we conclude from all this?

 

Based on the anecdotal reports of research subjects, it appears as if TB-500 is generally well tolerated with minimal chances of side effects appearing. But this is not definitive proof, so always use your best judgment when using TB-500 or any other therapeutic peptide.

TB-500 Dosage Guide

So what exactly is the RIGHT TB-500 dose to take for an athlete who wants to accelerate their rate of healing and recovery?

It depends.


Unlike medications which have been around for a long time, there is no universally agreed-upon dosing recommendation for TB-500.

Research subjects are left on their own to figure out what the optimal TB-500 dosage will be for their bodies.

One recommendation which seems to be consistent is taking two doses a week evenly spread throughout the week – Monday and Thursday, for instance.

2-4 mg of TB-500 administered per week seems to be the range in which suggested TB-500 dosing falls into.

Using the two points above, this is what would be considered a maintenance dose for someone who wants to heal an injury.

Some fitness health experts, such as Ben Greenfield, have recommended doses as high as 7-8 mg per week for someone who weights 200 pounds [17].

I even know “test subjects” who recommend a “loading” phase where you take 5 mg per week for 4-6 weeks before returning to a maintenance dose.
